Results showed a national prevalence of 18.11 % at the individual level and 85.93 % at the herd level. Significant variations in the individual prevalence were observed between regions of the country and ranged from 4.63 % in Tombouctou to 54.88 % in Kidal. At the herd level, although there were variations between regions, a high prevalence was constantly observed ranging from 60 to 100 %, hence confirming the endemic nature of the disease across the country. The CBPP risk varied also between regions and was very low in Tombouctou (odds ratio (OR) = 0.4) but very high in Kidal (OR = 9.8). Similarly, the risk seemed higher in the animals of the over 3-year age group (OR = 1.6) compared to the other age groups. It was also observed that there was a slightly higher risk (OR = 1.3) in the females than in the males. This study confirms the presence of CBPP across the country and should help to elaborate strategies for the effective control of the disease.

We show here that mycoplasmas-induced recall proliferation and IFN-γ responses are detected in cattle that received multiple shots of T1 vaccines. These anamnestic responses were under the strict control of CD4+ T lymphocytes. Moreover, CD62L expression indicated that both CD4+ effector memory (Tem) and central memory (Tcm) T lymphocytes are elicited in these animals. Comparative analysis with data from cattle that completely recovered from CBPP infection revealed similar anamnestic T-cell responses albeit at a lower magnitude for T1-vaccinated animals, particularly in the Tcm compartment. In conclusion, we discuss how our current understanding of T-cell responses will contribute to ongoing efforts for the improvement of future CBPP vaccines.

Thirty five clinically healthy zebu cattle, negative for CBPP antibodies (as tested by a competitive ELISA), were infected with Mycoplasma mycoides subsp. mycoidessmall colony variant - field isolate. All the animals were monitored for clinical signs of illness and regularly sampled for serological and bacteriological analysis. Nine animals were treated with long-acting tetracycline upon the first evidence of clinical signs of illness. The 26 remaining animals were left untreated and served as control animals. Post-mortem examinations were performed to assess the presence or absence of CBPP gross lesions. The experimentation lasted for 10 months during which, 13 animals (all untreated) died with acute lung lesions. When the 22 surviving animals (13 untreated and 9 treated) were slaughtered at the end of experimentation, chronic lesions were observed in all of them. Among the 13 untreated animals, 10 had visible lung sequestra and 3 had cicatrical lesions indicative of resolved lung lesions. Conversely, among the 9 treated animals, only 1 had a small lung sequestrum and the remaining 8 had pulmonary adhesions. The results presented demonstrate that, under the experimental conditions used, treatment with oxytetracycline did not result in significant sequestra formation in CBPP-infected animals. CBPP has been and is still a major cause for concern for African countries (due to mortality, animal-production losses and cost of monitoring and control). A cross sectional study of the herd and animal seroprevalence of Contagious Bovine Pleuropneumonia (CBPP) using stratified sampling; competitive cELISA test was carried out in a zebu cattle-producing district of Ethiopia, representative of the Ethiopian highlands fringe where CBPP is assumed to re-emerge after the phasing out in 1994 of Rinderpest and CBPP bivalent vaccination program. A total of 2508 zebu cattle (Horro breed) from 300 herds were examined serologically and for characteristic clinical signs of CBPP. The herd seroprevalence was 4.6% in average representing 15 herds. The herd seroprevalence was ranging from 2.0% to 8.5% in 3 strata representing increasing herd size. In conclusion, the clinical signs observed in the survey were not providing a good picture on the disease prevalence. (Resume d'auteur)

Time-Delay Dynamics for Contagious Bovine Pleuropneumonia

In this paper, we are interested in investigating how the distributions of sojourn times in infective compartments affect the dynamics and persistence of the Contagious Bovine Pleuropneumonia, a chronic respiratory disease of cattle. Two kinds of extreme distributions of the sojourn times are considered: a Dirac delta-function and truncated Gaussian function leading to a model with (non-constant) delay and the classical exponential distribution that stands for a model without delay. Expressions of the basic reproductive numbers are derived and dynamical behaviours are discussed for the three models. It is found that the spreading of disease exhibits wave-like oscillations for the time-delay dynamics. In contrast, the disease appears to last longer when the spreading is described by the classical dynamics without delay. Subsequently, the time-delay dynamics turns out to be more appropriate for the description of an experimental epidemic of CBPP.

Veterinary Journal, 2012Co-Authors: Yuan Li, Robin A J Nicholas, Chao Chen, Meijing Zhang, Hui DongAbstract:Abstract Contagious Bovine Pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides small colony type, was once the most damaging infectious animal disease in China, second only to rinderpest. Between 1949 and 1989, 178,570 cattle died of CBPP, causing estimated losses of 356 million RMB (1 RMB = approx. £0.094, US$0.15, €0.11 at 27th January 2011). In 1956, in an effort to control the disease, a virulent strain of the causative organism (Ben-1) was attenuated by multiple passages in rabbits. The resultant vaccine achieved a high protection rate in cattle with a duration of immunity of 28 months. Vaccines were also prepared in sheep to increase the antigen yield and then in Tibetan sheep because it caused fewer adverse reactions in yaks and related species. The last CBPP infected animal was identified in 1989 since when no more cases have occurred. In 1992, vaccination of cattle was stopped. In 2008 China submitted an application to the World Organization for Animal Health (OIE) to be declared CBPP-free.

Twenty isolates from various geographical locations and the type strain were analysed by multi-locus sequence analysis (MLSA). The data generated was then used to develop three PCR primer sets to differentiate these isolates. The PCRs differentiated the isolates into four groups; the type strain (T); isolates of European origin (Eu); isolates from Tanzania (Af1) with a final group consisting of isolates from Namibia and Botswana (Af2). These PCRs offers a rapid and efficient post-identification typing method without the need to sequence and analyse multiple genes.

Of the antimicrobials used most widely in Africa, oxytetracycline and tilmicosin were effective, while the isolates were resistant to tylosin. These results provide a baseline for monitoring antimicrobial resistance.

It is endemic in sub-Saharan Africa and causes important productivity losses due to the high mortality and morbidity rates. CBPP is caused by Mycoplasma mycoides subsp. mycoides (Mmm) and is characterized by severe fibrinous bronchopneumonia and pleural effusion during the acute to subacute stages and by pulmonary sequestra in chronic cases. Additional lesions can be detected in the kidneys and in the carpal and tarsal joints of calves. Mmm infection occurs through the inhalation of infected aerosol droplets. After the colonization of bronchioles and alveoli, Mmm invades blood and lymphatic vessels and causes vasculitis. Moreover, Mmm can be occasionally demonstrated in blood and in a variety of other tissues. In the lung, Mmm antigen is commonly detected on bronchiolar and alveolar epithelial cells, in lung phagocytic cells, within the wall of blood and lymphatic vessels, inside necrotic areas, and within tertiary lymphoid follicles. Mmm antigen can also be present in the cytoplasm of macrophages within lymph node sinuses, in the germinal center of lymphoid follicles, in glomerular endothelial cells, and in renal tubules. A complete pathological examination is of great value for a rapid presumptive diagnosis, but laboratory investigations are mandatory for definitive diagnosis. The purpose of this review is to describe the main features of CBPP including the causative agent, history, geographic distribution, epidemiology, clinical course, diagnosis, and control. A special focus is placed on gross and microscopic lesions in order to familiarize veterinarians with the pathology and pathogenesis of CBPP.

The peculiar pathological features of CBPP make desirable the assessment of ad hoc score methods to grade the disease in the affected animals. Thus, the present work aims to assess a new lung score system for CBPP. Our results indicate that the present score system strongly correlates with that previously published by Turner and could be effectively used in CBPP-affected animals.

The present study investigated the generation of ROS by different strains of Mmm , as well as their effect on the oxidative response of Bovine neutrophils. The production of ROS was indirectly measured using a luminol-based chemiluminescence assay. Our results confirm that Mmm can produce ROS via the metabolism of glycerol, significant differences existing between African and European strains. Mmm was capable of adhering to the external surface of neutrophils. Interestingly, Mmm enhanced the respiratory burst of Bovine neutrophils. This activity was particularly pronounced with the African field strain and in presence of glycerol. Taken together, our data argue in favour of a major role for neutrophils as the main source of ROS in Contagious Bovine Pleuropneumonia.
