Therefore, combination therapy with LLLT and extracts from the Guarana (Paullinia cupana) plant may improve its effectiveness as Guarana extracts exhibit anti-aging properties. OBJECTIVES To evaluate the antioxidant, anti-inflammatory, anti-apoptotic, and proliferative effects of combined LLLT and Guarana extract therapy on human dermal fibroblasts. METHODS Human dermal fibroblasts (HFF-1) were cultured and initially exposed to several concentrations (1, 3, 5, 10, 30 µg/mL) of Guarana extract. The experimental concentration of Guarana extract was selected by analyzing cytokine levels, DNA oxidation, and apoptotic markers in LLLT-exposed (4 J/cm2 ) and LLLT-unexposed fibroblast cultures. After 72 hours, the cells were analyzed using spectrophotometric, fluorimetric, immunological, and gene expression (qRT-PCR) assays. Flow cytometry was used to evaluate the effect of each treatment on cell cycle. RESULTS Fibroblasts treated with Guarana (5 µg/mL) exhibited anti-inflammatory and anti-apoptotic properties been used in complementary protocols. Combined Guarana and LLLT treatment significantly decreased protein carbonylation, lipoperoxidation, and DNA oxidation, downregulated the mRNA and protein expression of pro-inflammatory molecules, and upregulated IL-10 gene and protein expression. Guarana plus LLLT also decreased the levels of caspases 1, 3, and 8, increased the percentage of S-phase cells, and decreased FGF-1 and KGF-1 levels. Some of these changes were also observed after treatment with Guarana or LLLT alone. CONCLUSIONS Our results suggest that concomitant treatment with Guarana and LLLT may promote fibroblast biostimulation and thus is clinically relevant.

Western blot, clonogenic assay, and growth curve were performed. Moreover, we investigated the potential cytotoxic effect of Guarana in normal cells. Materials and methods: Male Wistar rats were pretreated with Guarana powder (100, 300 and 600 mg/kg) or silymarin 100 mg/kg daily for 14 days before treatment with a single dose of CCl4 (50% CCl4, 1 mL/kg, intraperitoneally). Results: The treatment with CCl4 significantly increased the serum activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). In vitro supplementation by antioxidant food extract of senescent mesenchymal stem cells could reverse these undesirable characteristics. To evaluate this hypothesis, senescent adipocyte-mesenchymal cells (ASCs) obtained from human lipoaspirates were exposed at different concentrations of hydro-alcoholic Guarana ( Paullinia cupana ) extract for 72 h. After the incubation, we performed a proliferative assay. Oxidative stress indicators and antioxidant enzymes (biochemical activity and gene expression by qRT-PCR analysis) in these senescent cells were also evaluated. In senescent cells exposed to Guarana at 5 mg/g concentration increased cellular proliferation occurred compared to untreated senescent cells (79.1 ± 15.7%). Concomitantly, a decrease in several oxidative stress indicators was observed in senescent cells treated with Guarana. A genomic effect of Guarana exposure was observed when the modulation of antioxidant enzymes genes was analyzed. The results described here suggest that the food extract supplementation could reverse the initial senescence processes in ASCs. These results have potential application in regenerative medicine.

Given that nutrition may influence some age-related disorders, this study investigated, in vivo, the association between self-reported eyesight quality and daily Guarana intake in riparian elderly subjects and, in vitro, the effects of Guarana (Paullinia cupana) on retinal pigmented epithelium (RPE) cells exposed to paraquat. Just over half of the elderly subjects self-reported poor vision (n = 320, 50.8%), 234 (37.1%) self-reported regular eyesight, whereas just 76 (12.1%) self-reported good vision. Subjects that self-reported good vision presented a higher frequency of habitual Guarana consumption. Guarana treatment was able to decrease apoptosis and necrosis events and differentially modulate gene expression and protein levels of caspases 1, 3 and 8 in RPE cells. This work provides new perspectives on the potential use of Guarana against eye diseases due to its cytoprotective effects.

First, cells were exposed to VCR (0.009 µM for 24 hours and 0.0007 µM for 72 hours) to measure the cytotoxicity effect. Also, the cellular effect of hydroalcoholic extract of Guarana (10; 30; 100 and 300 μg/mL) was evaluated in the same cells in 24 and 72 hours. After that, cells were exposed to VCR and Guarana extract to evaluate the neuroprotective effect of Guarana. Measurements Cell viability was analyzed by MTT, Free dsDNA and LHD Assays. Moreover, metabolism oxidative profile was evaluated by reactive oxygen species (ROS), lipoperoxidation (LPO) and catalase (CAT) levels through DCFH-DA, TBARS and Catalase Activity Assays, respectively. Results Our findings revealed that VCR caused neuronal cytotoxicity by reducing cell viability and increasing ROS and LPO levels. On the other hand, Guarana did not cause cell damage in none of tested concentrations. In addition, Guarana exhibited a notable protective effect on brain and cerebellum cells exposed to VCR by increasing cell viability, stimulating CAT activity, reducing levels of ROS and LPO. Conclusions In this sense, Guarana is a remarkable antioxidant fruit that could be a target in new therapies development to reduce VCR neurotoxicity. .

Worsening of Oxidative Stress, DNA Damage, and Atherosclerotic Lesions in Aged LDLr-/- Mice after Consumption of Guarana Soft Drinks.

VasquezAbstract:Background. Excessive consumption of soft drinks (SD) has become a health problem worldwide due to its association with related cardiovascular diseases. We investigated the possible impacts associated with the consumption of Brazilian Guarana (normal and zero) SD in dyslipidemic mice, thus mitigating potential clinical confounders such as poor-quality diet, lifestyle, body composition, and/or comorbidities. Methods. Sixteen-month-old LDLr-/- mice were divided into the following groups: (1) control; (2) GSD: normal Guarana SD; and (3) Z-GSD: zero Guarana SD. All were fed ad libitum, and blood pressure was measured noninvasively. After 8 weeks, aorta, blood, liver, and stomach samples were collected for histological and biochemical analyses. Results. Guarana soft drinks increased atherosclerosis (~60%) and were associated with hypercholesterolemia, hypertension, oxidative stress, DNA fragmentation, and apoptosis (~2-fold) of blood cells, besides presenting an increase in liver and gastric damage even in normoglycemia. Interestingly, Z-GSD did not cause the aforementioned changes, except in hemodynamic and renal parameters. Conclusions. Chronic administration of GSD is prooxidative, compromising the cardiovascular, gastric, and hepatic systems; the effects are due at least in part to free sugar consumption but not to Guarana extract per se.

Guarana (Paullinia cupana): toxic behavioral effects in laboratory animals and antioxidant activity in vitro

Dias, Silvia Berlanga De Moraes BarrosAbstract:The effects on toxic and behavioral levels of Guarana (Paullinia cupana) were assessed in rats and mice subsequent to acute and chronic administrations and were compared to those produced by Ginseng (Panax ginseng). Experimental parameters included tests for antioxidant capacity in vitro and measured in vivo, toxicological screening, progress in weight, motor activity, death rate, and histopathological examination of the viscera. Guarana showed an antioxidant effect because, even at low concentrations (1.2 microg/ml), it inhibited the process of lipid peroxidation. In high doses of 1000-2000 mg/kg (i.p. and p.o.) it did not induce significant alterations in parameters for toxicological screening. No effects on motor activity were observed, neither did Guarana alter the hypnotic effect of pentobarbital. Ginseng (250-1000 mg/kg i.p.), however, elicited reductions in motor activity, eyelid ptosis and bristling fur. Consumption of liquids containing Guarana or ginseng and progress in weight of the animals remained at levels similar to the controls, even after prolonged administration. The percentage mortality was equivalent in control and in treated groups. The absence of toxicity of Guarana was also demonstrated by histopathological examination, with no alteration being detected in heart, lungs, stomach, small and large intestine, liver, pancreas, kidneys, bladder and spleen.

Pharmacological activity of Guarana (Paullinia cupana Mart.) in laboratory animals

Dias, Rita Mattei, Elisaldo Araújo CarliniAbstract:Mice that ingested a suspension of Guarana (Paullinia cupana, Sapindaceae) in a dose of 0.3 mg/ml showed a significant increase in physical capacity when subjected to a stressful situation such as forced swimming after 100 and 200 days of treatment. Such an effect, however, was not obtained with a concentration of 3.0 mg/ml, nor with the ingestion of a suspension of ginseng 5.0 mg/ml, nor of a solution of caffeine 0.1 mg/ml. Guarana, both after a single (3.0 and 30 mg/kg) or chronic administrations (0.3 mg/ml), was able to partially reverse the amnesic effect of scopolamine as measured through a passive avoidance test in mice and rats, indicating a positive effect on memory acquisition. However, no effect was observed when an active avoidance task was used in rats, even after 20 days of Guarana administration. There was also a tendency of rats treated with 0.3 mg/ml of Guarana to better maintain the memory of a Lashley III maze path. The animals had the same average lifespan, indicating a low toxicity of Guarana, even after 23 months of treatment.

The authors assessed the effects of the long-term administration of Guarana on the cognition of normal, elderly volunteers. Forty-five volunteers were studied, with a random distribution in three experimental groups: placebo (n = 15), caffeine (n = 15), and Guarana (n = 15), in a double-blind study. There were no significant cognitive alterations in these volunteers.

Although results were negative it cannot be concluded that "Guarana" is harmless. Other studies shall be undertaken, administering "Guarana" on a long term basis, as popularly proclaimed.
