The fish in the infected group were challenged with gill monogeneans by cohabitation method. The control group fish were not exposed to parasites. Susceptibility and pathological consequences of Catla, Catla Catla (Hamilton) experimentally infected with Edwardsiella tarda

Jawahar Abraham, Dibyendu KamilyaAbstract:AbstractThe present study tested the susceptibility and pathological changes of Catla, Catla Catla (Hamilton) infected with Edwardsiella tarda (ET-PG-29). The bacterium was isolated from the kidney of a diseased pangas catfish. To determine the median lethal dose (LD50), C. Catla were challenged with this bacterium (108-103CFU ml-1), and the LD50was calculated as 105.5CFU ml-1. Another set of healthy C. Catla were injected intraperitoneally with the LD50dose to induce edwardsiellosis. The clinical signs of the infected C. Catla were observed and recorded. Tissues such as kidney, liver, intestine, heart, and gill from the infected fish with clinical signs of edwardsiellosis were used for histopathology. The clinical and gross signs were first visible at 1 d post-injection, and the infected fish showed typical signs of hemorrhagic septicemia. The most striking histopathological features were found in the kidney which showed multi-focal necrosis with the formation of granuloma indicating an inflammatory response against the pathogen. The intestine displayed goblet cell hyperplasia, the liver showed hydropic degeneration with hyperemic central veins, and there was inflammation of gill lamellae and cardiac myositis associated with leucocyte infiltration. Collectively, the results confirmed the susceptibility of C. Catla to E. tarda infection and that this bacterium is a threat to C. Catla in aquaculture practices.

The probiotic attributes of B. subtilis was tested by conducting antagonism study, safety in Catla, in vitro immunomodulation and dietary immunomodulation. Results of these studies indicated the probiotic potential of the strain. From the preliminary dietary immunomodulation study, a dose of 10(9) B. subtilis cells g(-1) was selected for inclusion into diets for subsequent experiments. Experimental diets were prepared by adding B. subtilis (10(9) cells g(-1)), chitin (2%) and their combination to the basal diet. Different systemic and mucosal immunological parameters viz. oxygen radical production, myeloperoxidase content, lysozyme activity, total protein content and alkaline phosphatase activity showed significant enhancement (p<0.05) after 2 weeks of feeding with the combined diet. B. subtilis and chitin alone also significantly elevated most of the immune responses. All the diets significantly increased the resistance of Catla against E. tarda challenge. The highest post-challenge survival was observed in combined group (i.e. 63.33%). In conclusion, B. subtilis and chitin, alone or combined, had a health ameliorating effect in Catla. The results also collectively suggest the usefulness of applying a combined probiotic and immunostimulant supplemented diet to achieve greater benefits.

Achyranthes has significantly (P<0.05) enhanced the BSA-specific antibody titers than the untreated control group throughout the study period. The efficiency of antigen clearance was also enhanced in C. Catla treated with Achyranthes.

Effects of UV-B radiation on the gills of Catla Catla during early development

Fish without UV-B exposure served as control. UV-B radiation damaged both gill filaments and lamellae. The intensity of damage was minimal in 5 min exposed fish, followed by 10 min exposed fish and maximal in 15 min. The gill epithelium was severely damaged in 15 min irradiated fish compared to control. Pavement cells (PVCs) were damaged and the numbers of microridges within PVCs decreased. The deep boundary of PVC was not clear. In some area of gill epithelium, PVCs were destroyed and mitochondrion-rich cells (MRCs) were exposed. The 5 min exposure reduced the number of microridges in the PVCs, but the boundary of PVCs was still visible. MRCs in the gill epithelium were not exposed in 5 min exposed fish. Catla head kidney leukocytes (HK) were incubated with the bacteria alive or inactivated with heat shock (2 h at 60 °C), UV light (for 2.5 h) and formalin (1.0 %, v/v) treatment (for 24 h at 4 °C) at different concentrations (10^7, 10^8 and 10^9 cells ml^−1). After incubation, different cellular immune parameters such as respiratory burst activity, nitric oxide production, leukocyte peroxidase content and proliferative response were analyzed. The inactivated probiotic preparations stimulated all the cellular immune parameters of Catla HK leukocytes in vitro. Among the different inactivated preparations, heat-treated form exhibited the best result. The lowest dose (10^7 cells ml^−1) of both the strains showed the maximum stimulation. The results collectively suggest the efficacy of inactivated preparations to be used as immunostimulant in aquaculture.

Larvae were fed with exogenous live plankton. Cultures at 6,667 and 8,333 larvae/m3 showed significantly (P <0.05) higher survival and growth than larvae stocked at 10,000 larvae/m3 for both species. Food was more efficiently used in low stocking density, as evident from the significantly (P <0.05) lower values of feed conversion ratio in lower density compared to those for high stocking density. Specific growth rate of both species was high in the early stage and gradually declined along with the ontogenic development. Development and characterization of cell lines derived from rohu, Labeo rohita (Hamilton), and Catla, Catla Catla (Hamilton)

Sahul HameedAbstract:Two new cell lines, designated RE and CB, were derived from the eye of rohu, Labeo rohita, and the brain of Catla, Catla Catla, respectively. The cell lines were maintained in Leibovitz's L-15 supplemented with 20% foetal bovine serum. The RE cell line was sub-cultured for more than 70 passages and the CB cell line for more than 35 passages. The RE cells are rounded and consist predominantly of epithelial cells. The CB cell line consists of predominantly fibroblastic-like cells. Both cell lines are able to grow at temperatures between 25 and 32 degrees C with an optimum of 28 degrees C. The growth rate of the cells increased as the foetal bovine serum concentration increased from 2% to 20% at 28 degrees C, with optimum growth at concentrations of 15% or 20% foetal bovine serum. The cells were successfully cryopreserved and revived at different passage levels. The cell lines were not susceptible to four marine fish viruses. Extracellular products from Aeromonas sp. were toxic to the cell lines. When the cells were transfected with plasmid eukaryotic green fluorescent protein (pEGFP [Clontech, Carlsbad, CA, USA]) vector DNA, a significant fluorescent signal was observed suggesting that these cell lines could be a useful tool for transgenic and genetic manipulation studies. Polymerase chain reaction amplification of mitochondrial 12S rRNA from rohu and Catla confirmed that the cell lines originated from these fish species. The cell lines were further characterized by immunocytochemistry using confocal laser scanning microscopy.
