    Abstract:

    Black mulberry is a widely acknowledged ancient traditional medicine. Its extract and constituents have been reported to exert various bioactivities including antimicrobial, hypotensive, analgesic etc. effects. While black mulberry preparations are also used as antispasmodic agents in folk medicine, no related studies are available on its isolated constituents. Through an extensive chromatographic purification, seven phenolic compounds were isolated from the methanol extract of Morus Nigra root bark, including Morusin (1), kuwanon U (2), kuwanon E (3), moracin P (4), moracin O (5), albanol A (6), and albanol B (7). A complete NMR signal assignment of moracin P and O was achieved, and related literature errors confusing the identity of moracin derivatives are hereby clarified. Compounds 2, 5 and 7 were identified as strong antispasmodic agents on isolated rat ileum and tracheal smooth muscles, while compound 3, a methoxy derivative of 2, was inactive. Moracin O (5) inhibited the ileal and tracheal smooth muscle contractions with Emax values of 85% and 302 mg, respectively. Those actions were superior as compared with papaverine. Our findings demonstrate that prenylated arylbenzofurans, geranylated flavonoids and Diels-Alder adducts from Morus Nigra are valuable antispasmodic agents. Compounds 2, 5 and 7 are suggested as marker compounds for quality control of antispasmodic mulberry preparations. Moracin O (5) is a new lead compound for related drug development initiatives.

    Abstract:

    Phenolic compounds are of considerable biomedical interest due to their antioxidant properties and potential in the prevention and possibly treatment of many chronic diseases. The fruits, leaves and root bark of Morus Nigra (Moraceae), the black mulberry tree, have a long history of use for various therapeutic purposes in traditional medicine worldwide. The roots of the plant are known to be a rich source of phenolic compounds with a particularly high chemical diversity. This mini-review compiles the currently available knowledge on phenolic compounds reported from Morus Nigra roots, and provides a brief overview on the antioxidant activity with a focus on the available in vivo evidence. Morusin, a major phenolic antioxidant of the root bark, has attracted a rapidly increasing scientific interest for its versatile and potent antitumor properties; recent developments in this regard, including Morusin’s promising activity against cancer stem cells, are also discussed in the paper.

    Este estudo teve como objetivo analisar o potencial inovador da planta medicinal Morus Nigra L. a partir  do numero de patentes depositadas nos bancos de dados European Patent Office (EPO), World Intellectual Property Organization (WIPO) e Instituto Nacional de Propriedade Industrial do Brasil (INPI). Morus Nigra L. e conhecida popularmente como “amora-miura”, sendo bastante utilizada para o tratamento de diabetes, dislipidemia, problemas cardiovasculares e gota. A pesquisa foi realizada utilizando como descritores os termos: “Morus Nigra”, “medicament” ou “medicamento” e “food supplement” ou “suplemento alimentar”. Os resultados demonstraram um  elevado numero de documentos envolvendo o termo “Morus Nigra”, com predominância para o World Intellectual Property Organization (24) em comparacao ao European Patent Office (23) e ao INPI (4). No entanto, combinando os descritores “Morus Nigra and medicament” esse numero decresceu (WIPO – 8; EPO – 3; INPI – 0). Considerando os dados coletados, verificou-se que a Organizacao Mundial de Propriedade Intelectual e a maior detentora de numero de patentes, com destaque para os depositos nas subclasses da Classificacao Internacional de Patentes em A61K e A61P, em ambos os bancos tecnologicos. A Morus Nigra apresenta potencial inovador considerando-se as atividades farmacologicas atribuidas a esta especie e as patentes ate o momento depositadas no WIPO, EPO e INPI.

    Abstract:

    Morus Nigra L. is known in the region of Brazilian Sao Francisco Valley as "amora-miura". It is widely used in traditional medicine, mainly in treatment of diabetes, hypercholesterolemia, cardiovascular problems, obesity and gout. This study proposed to develop and validate a fast, simple, efficient and low cost analytical method that could quantify total flavonoid content present in crude ethanolic extract of leaves of M. Nigra (Mn EtOH). For this, ultraviolet-visible (UV-Vis) absorption spectrophotometry was used as the analytical tool and the complexation with aluminium chloride (AlCl3). The quantification of flavonoids by this method was based on the complexation of AlCl3 with the flavonoid nucleus as a selectivity tool made by two procedures: without complexation (NCP) and with complexation (WCP), in order to evaluate the effects of the complexing agent in quantification, proving the efficiency of this technique. In this way, it became necessary to validate the method used to ensure its efficiency. The validation of the method of quantification of total flavonoid content by UV-Vis demonstrated that the method was selective, linear, precise, accurate, and robust.   • Acaricidal activity of leaves of Morus Nigra against the cattle tick Rhipicephalus microplus

    Abstract:

    ABSTRACT The acaricidal activity of crude ethanolic extract and fractions from the leaves of Morus Nigra (Moraceae) was carried out on female cattle ticks Rhipicephalus microplus, using the adult immersion test. The mortality and fertility of females exposed to different concentrations of hexane, chloroform and ethyl acetate fractions, as well as ethanolic extract of M. Nigra with concentrations of 5, 10 and 25mg/ml were evaluated using three treatment groups, two control groups and triplicate tests. The study also identified the main phenolic compounds of the extract and fractions of this species by HPLC. The chloroform fraction of leaves of M. Nigra (25mg/mL) showed the best results for this species, obtaining 62.6% of inhibition of oviposition, 39.3% of eggs eclosion average and 65.4% of effectiveness. HPLC analysis revealed the presence of phenolic compounds, which may be related to biological activity shown by the extracts, which can be used as an alternative control against R. microplus adult tick.

    Abstract:

    Morus Nigra has been used popularly for several proposes, including diabetic. In an attempt to support medicinal value, the acute hypoglycemic, hypolipidemic, and antioxidant effects of the ethanolic extract of Morus Nigra (EEMn 200 or 400 mg/kg b.w.) were evaluated in normal and alloxan-induced diabetic treated for 14 days. Serum biochemical and antioxidant analysis were performed at the end of experiment. Oral glucose tolerance test was performed at 10th and 15th days. Chromatographic analysis by HPLC-DAD of EEMn was performed. Insulin was used as positive control to glycemic metabolism as well as fenofibrate to lipid metabolism. EEMn (400 mg/kg/day) reduced fasting and postprandial glycaemia, improved oral glucose tolerance, and reduced lipolysis and proteolysis in diabetic rats. EEMn decreased the blood levels of total cholesterol and increased HDL level when compared to the diabetic control rats. At higher levels, EEMn reduced triglycerides and VLDL levels in diabetic rats. Also, EEMn reduced malondialdehyde and increased the reduced glutathione levels in liver of diabetic rats. Chromatographic analysis identified the presence of the flavonoids rutin, isoquercetin, and kaempferitrin. Acute EEMn treatment reduced hyperglycemia, improved oral glucose tolerance, and minimized dyslipidemia and oxidative stress leading to a reduction in atherogenic index in alloxan-induced diabetic rats.

    Investigate the anti-irradiation effect of Morus Nigra L.pigment on zebrafish embryo.Methods 4h post fertilization zebrafish embryos were treated with various concentrations(0.5,5,50ug / mL) Morus Nigra L.pigment.Three hours later,x-ray(20Gy) was applied.Then the level of ROS in zebrafish embryo was determined by ROS probe(2,7-dichloro fluorescein diactate,DCFH-DA),and the content of superoxide dismutase(SOD) and malondialdehyde(MDA) in the zebrafish embryo tissue were evaluated by biochemical method assay.Results compared with the control and irradiation group,ROS content was significantly different(p 0.01) with different concentration of pigment;The activity of SOD was not obvious increased(P 0.05);In 5 and 50μg / mL groups,the content of MDA reduced with the increase of pigment concentration and was significantly lower compared with the control gropu(p 0.01).Conclusion Morus Nigra L.pigment had radiation protection ability of zebrafish embryos,the mechanism may due to the improvement the SOD activity,reducing oxygen free radicals and inhibiting the lipid peroxidation.The test also showed that Morus Nigra L.pigment had anti-oxidative activity.

    The total flavonoids in the tree branch bark of Xinjiang black mulberry(Morus Nigra L.) were obtained by reflux extraction method.The total content of flavonoids was measured by UV-spectrophotometric method.The optimal extraction conditions were investigated through single factor and orthogonal experiments.The optimal conditions were as followed: extraction temperature 80℃,solid liquid ratio 1∶50,extraction time 120min and ethanol concentration 70%vol.Under the optimal extraction conditions,the extraction efficiency of total flavonoids was 32.65mg/g,which indicated that this method was feasible and reasonable.

    A new 2-arylbenzofuran derivative, mornigrol D (1), along with two new flavones, mornigrol G (2) and mornigrol H (3), and six known compounds, norartocarpetin (4), dihydrokaempferol (5), albanin A (6), albanin E (7), moracin M (8), and albafuran C (9), were isolated from the barks of Morus Nigra. Their structures were elucidated by spectroscopic analysis. Compounds 1 and 9 showed antioxidative activities in vitro with inhibition ratios of 98 and 99% at the concentration of 10− 4 mol/l, and of 74 and 75% at the concentration of 10− 5 mol/l. In addition, compounds 1 and 4 showed potent anti-inflammatory activities (inhibition of release of β-glucuronidase from rat polymorphonuclear leucocytes induced by platelet activating factor) with inhibitory ratios of 65.9% (P < 0.01) and 67.7% (P < 0.01) at a concentration of 10− 5 mol/l.

    OBJECTIVE To investigate the chemical constituents in the barks of Morus Nigra. METHOD The nine compounds were isolated and purified by column chromatography over silica gel, LH -20 and preparative HPLC and their structures were identified by various spectroscopic methods. RESULT The nine compounds were isolated and identified as olcancolic acid , apigenin, cyclocommunol, Morusin, cycloMorusin, kuwanon C, daucosterol, ursolic acid, 63-sitosterol. CONCLUSION Olcancolic acid, apingenin, cyclocommunol, Morusin, cycloMorusin, kuwanon C, daucosterol were firstly isolated from the M. Nigra.
