    As part of on-going efforts to control hookworm infection, the “human hookworm vaccine initiative” has recognised blood feeding as a feasible therapeutic target for inducing immunity against hookworm infection. To this end, molecular approaches have been used to identify candidate targets, such as Necator americanus (Na) haemoglobinase aspartic protease-1 (APR-1), with immunogenicity profiled in canine and hamster models. We sought to accelerate the immune analysis of these identified therapeutic targets by developing an appropriate mouse model. Here we demonstrate that Nippostrongylus brasiliensis (Nb), a phylogenetically distant strongylid nematode of rodents, begins blood feeding early in its development and that immunisation with Na-APR-1 can block its growth and completion of its life cycle. Furthermore, we identify a new haem detoxification pathway in Nb required for blood feeding that can be blocked by drugs of the quinolone family, reducing both infection burden and the associated anaemia in rodents. Collectively, our findings show that haem metabolism has potential as a checkpoint for interrupting hookworm development in early stages of the hookworm life cycle and that the Nippostrongylus brasiliensis rodent model is relevant for identifying novel therapeutic targets against human hookworm.

    Abstract:

    Nippostrongylus brasiliensis , a nematode parasite of rodents, has a parasitic life cycle that is an extremely useful model for the study of human hookworm infection, particularly in regards to the induced immune response. The current reference genome for this parasite is highly fragmented with minimal annotation, but new advances in long-read sequencing suggest that a more complete and annotated assembly should be an achievable goal. We de-novo  assembled a single contig mitochondrial genome from N. brasiliensis  using MinION R9 nanopore data. The assembly was error-corrected using existing Illumina HiSeq reads, and annotated in full (i.e. gene boundary definitions without substantial gaps) by comparing with annotated genomes from similar parasite relatives. The mitochondrial genome has also been annotated with a preliminary electrical consensus sequence, using raw signal data generated from a Nanopore R9 flow cell.

In particular, they model the induction and maintenance of Th2 type immune responses and exhibit all the requisite hallmarks of CD4 T cell-dependent IgE production, eosinophilia, mastocytosis, and mucus production. This chapter describes simple, cost-effective techniques for using and maintaining these easy-to-work-with parasites in the context of a modern laboratory.

  • Influence of Sample Recovery Techniques on Detection of Biogenic Amines in the Rat Hookworm Nippostrongylus brasiliensis

    Abstract Biogenic amines in Nematoda have previously been found by means of high-performance liquid chromatography (HPLC) equipped with various detectors. The methods of sample preparation have differed widely. In this study, we tested the influence of experimental conditions (temperature and duration of exposure to light, centrifugation or individual recovery by pipetting each worm, and saccharose gradient versus physiological saline) on catecholamines and indolamines content in Nippostrongylus brasiliensis at various developmental stages (larvae and adults). Amines were determined by liquid chromatography with electrochemical detection. To the best of our knowledge, octopamine, tyramin, N-acetylserotonin and N-acetyldopamine were found and determined for the first time in this nematode. All experimental conditions listed above modify the endogenous amine levels in larval and adult Nippostrongylus brasiliensis. This should be taken into account in the design of future research protocols.

    Microtopographic features of the various growth stages of the three free-living larval stages of the rat hookworm Nippostrongylus brasiliensis (Nematoda) were surveyed by scanning electron microscopy. These worms have a rounded anterior end and an elongated tail. Cuticular annulations were observed along the body, which also bore two ribbon-like lateral alae. Two rings of six lip-like lappets were observed around the triradiate oral opening in all larval stages. The cephalic space contained two lateral amphidial pits. The excretory pore in the third anterior part was observed in a ventral view of the larvae. No deirids were observed. The anus with a crescent-shape opening was located posteriorly. Phasmidial apertures, only observed in the third-stage larvae, opened on the lateral alae in the tail region. © 1993 Wiley-Liss, Inc.

    Abstract 1. The influence of ecdysteroids (ecdysone and ecdysterone) was investigated on the control of reproduction in the parasitic nematode Nippostrongylus brasiliensis in vivo. 2. Infestive larvae (L3) were immersed in solutions of ecdysteroids (2.2 μM) at 37°C for 4 hr before injection into the host. The effect on egg-laying was observed two stages later. 3. The treatment increased egg-laying, but had no influence on the timing of the reproductive period. The greatest effect was observed with ecdysone, ecdysterone only inducing a small and non-significant stimulation under our experimental conditions. 4. The physiological role of ecdysteroids in meiosis and gonadal development in nematodes is discussed.

    Abstract 1. Simultaneous detection (HPLC and electrochemical detection) of biological extracts of larval and adult stages of Nippostrongylus brasiliensis was performed in order to assay biogenic amines. 2. Gamma-amino-butyric acid was assayed in the same samples. 3. Tryptophan, 5-hydroxyindoleacetic acid were at the same level in adults and larvae. 4. 5-Hydroxytryptophan, serotonin, dihydroxyphenylalanine and dopamine were significantly higher in larvae in which gamma-amino-butyric acid was not detected.
